mRNA expression analysis
Strain specific analysis of CD112 gene expression in wild-type C57BL/6 mice and B-hCD155/hCD112 mice by RT-PCR. Mouse Cd112 mRNA was detectable only in colon of wild-type C57BL/6 mice (+/+). Human CD112 mRNA was detectable only in homozygous B-hCD155/hCD112 mice (H/H;H/H), but not in wild-type mice.
Protein expression analysis
Strain specific CD155 expression analysis in heterozygous B-hCD155/hCD112 mice by flow cytometry. Dendritic cells of spleen and macrophages of peritoneal lavage fluid were collected from wild-type C57BL/6 mice (+/+) and heterozygous B-hCD155/hCD112 mice (H/+;H/+), and analyzed by flow cytometry with species-specific anti-CD155 antibody. Mouse CD155 was detectable in wild-type mice and heterozygous B-hCD155/hCD112 mice. Human CD155 was exclusively detectable in heterozygous B-hCD155/hCD112 mice but not in wild-type mice.

Mouse CD112 was detectable in wild-type mice. Human CD112 was exclusively detectable in heterozygous B-hCD155/hCD112 mice but not in wild-type mice. Anti-mCD112 antibody is a human-mouse cross-binding antibody.
