    BCG014: A CDH3-Targeting ADC with a Novel TOPO1 Inhibitor Payload

    CDH3 has been identified as a widely overexpressed antigen in various cancers. We utilized RenLite CDH3 knockout mice to identify Ab.20, which specifically targets CDH3. Ab.20 effectively binds to multiple tumor cell lines and demonstrates strong internalization activity. In the proof-of-concept study, the Ab.20-vcMMAE showed superior efficacy compared to the benchmark Ab-vcMMAE in PDX models. Additionally, Ab.20 was conjugated with BLD1102, a novel topoisomerase I inhibitor payload, forming BCG014, which demonstrated potent efficacy in a PDX model. Ongoing studies are designed to further evaluate the efficacy, PK, and toxicity of BCG014.
