      Description
      • IL32 (interleukin 32), is a member of the cytokine family. The encoded protein contains a tyrosine sulfation site, 3 potential N-myristoylation sites, multiple putative phosphorylation sites, and an RGD cell-attachment sequence. Expression of this protein is increased after the activation of T-cells by mitogens or the activation of NK cells by IL-2. This protein induces the production of TNFalpha from macrophage cells.
      • The full coding sequences of human IL32 gene, including the promoter, 5’UTR and 3’UTR are inserted into mouse Hipp11 (H11) locus in B-hIL32 mice.
      • Human IL32 mRNA was detectable in homozygous B-hIL32 mice.
      • Human IL32 was only detectable in the cell lysate of homozygous humanized B-hIL32 mice but not in wild-type C57BL/6JNifdc  mice, and increased after LPS stimulation .
      • Application: This product is used for pharmacological and safety evaluation of antibody drugs.

      Gene targeting strategy for B-hIL32 mice. The full coding sequences of human IL32 gene, including the promoter, 5’UTR and 3’UTR are inserted into mouse Hipp11 (H11) locus in B-hIL32 mice.

      mRNA expression analysis

      Strain specific analysis of IL32 mRNA expression in wild-type C57BL/6JNifdc mice and B-hIL32 mice by RT-PCR. Spleen RNA was isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hIL32 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with human IL32 primers. Human IL32 mRNA was exclusively detectable in homozygous B-hIL32 mice  mice but not in wild-type mice.

      Protein expression analysis in cell lysate

      Strain specific IL32 expression analysis in wild-type C57BL/6JNifdc mice and homozygous B-hIL32 mice by ELISA. Splenocytes were collected from wild-type C57BL/6JNifdc(+/+) and homozygous B-hIL32 mice (H/H) (n=3), then stimulated by 1 μg/ml LPS in cell culture for 18 h. Cell lysates were collected after stimulation. Expression level of human IL32 were analyzed by ELISA (Human IL32 ELISA Kit : RD , DY3040-05). Human IL32 was exclusively detectable in cell lysate of homozygous B-hIL32 mice with or without stimulation. Human IL32 expression was increased after LPS stimulation. Values are expressed as mean ± SEM.
