Description
CNTFR: A Crucial Role in Neuronal Development, Maintenance, and Survival
- Gene Information: Ciliary neurotrophic factor receptor (CNTFR) is a protein-coding gene encoding the CNTFRα subunit, a glycosylphosphatidylinositol (GPI)-anchored receptor component of the cytokine receptor family. CNTFRα mediates signaling of ciliary neurotrophic factor (CNTF) and related cytokines.
- Protein Expression: CNTFRα is expressed predominantly in the central nervous system, including hypothalamic and neuronal populations, and is also expressed in peripheral tissues.
- Signaling Pathway: The activation of CNTFR triggers a cascade of intracellular signaling pathways that influence cellular processes such as survival, differentiation, and apoptosis (programmed cell death). Specifically, when CNTFR is activated by its agonists, it often involves the activation of the JAK/STAT (Janus kinase/signal transducer and activator of transcription) pathway, as well as the MAPK (mitogen-activated protein kinase) pathway.
- Therapeutic Agonists: CNTFR agonists are currently being investigated for a range of therapeutic applications, primarily within the realm of neurodegenerative diseases. One of the most promising areas of research is their use in treating conditions such as amyotrophic lateral sclerosis (ALS), multiple sclerosis (MS), and spinal cord injuries.
Targeting strategy
CNTFR
- The exons 1-10 of mouse Cntfr gene that encode the whole molecule, including promoter, 5’UTR, and 3’UTR, were replaced by human counterparts in B-hCNTFR mice.
- The human CNTFR expression is driven by the human CNTFR promoter.
- The mouse Cntfr gene transcription and translation will be disrupted.
CNTFR mRNA Expression Analysis
Strain specific analysis of CNTFR mRNA expression in wild-type C57BL/6Nifdc mice and B-hCNTFR mice by RT-PCR. Brain RNA were isolated from wild-type C57BL/6Nifdc mice (+/+) and homozygous B-hCNTFR mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CNTFR primers. Mouse Cntfr mRNA was only detectable in wild-type mice. Human CNTFR mRNA was exclusively detectable in homozygous B-hCNTFR mice but not in wild-type mice.
