      Description
      • Kv1.3, identified as a predominant potassium channel in TEM cells, is upregulated during T-cell activation. Its tissue-specific overexpression in TEM subsets is critical for sustaining the membrane potential necessary to mediate inflammatory cascades.
      • The exon 1 of rat Kcna3 gene that the full-length protein was replaced by human counterpart gene sequences in B-hKCNA3 rats. The promoter, 5’UTR and 3’UTR region of the mouse Kcna3 are also retained. The human KCNA3 expression is driven by endogenous rat Kcna3 promoter, while rat Kcna3 gene transcription and translation will be disrupted.
      • KCNA3 was detectable in brain, lung, thymus, spleen and stomach of wild-type SD rats and homozygous B-hKCNA3 rats.
      • Application: This product is used for preclinical safety evaluation of antibody drugs.
      Targeting strategy

      Gene targeting strategy for B-hKCNA3 rats. The exon 1 of rat Kcna3 gene that the full-length protein was replaced by human counterpart gene sequences in B-hKCNA3 rats. The promoter, 5’UTR and 3’UTR region of the mouse Kcna3 are also retained. The human KCNA3 expression is driven by endogenous rat Kcna3 promoter, while rat Kcna3 gene transcription and translation will be disrupted.

      mRNA expression analysis

      Strain specific analysis of KCNA3 mRNA expression in wild-type SD rats and homozygous B-hKCNA3 rats by RT-PCR. Brain RNA was isolated from wild-type SD rats (+/+) and homozygous B-hKCNA3 rats (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with rat or human KCNA3 primers. Rat Kcna3 mRNA were detectable in wild-type rats. Human KCNA3 mRNA was detectable only in homozygous B-hKCNA3 rats but not in wild-type rats.

      Protein expression analysis

      Strain specific KCNA3 expression analysis in homozygous B-hKCNA3 rats by western blot. Brain, lung, thymus, spleen and stomach were collected from SD rats and homozygous B-hKCNA3 rats (H/H) and analyzed by western blot with KCNA3 (Proteintech, 14079-1-AP). KCNA3 was detectable in brain, lung, thymus, spleen and stomach of wild-type SD rats and homozygous B-hKCNA3 rats. The KCNA3 antibody cross-reacts with both human and rat.
