Description
DR4 is 56 kD member 10A of the TNFR superfamily (TNFRSF10A), also known as TRAIL-R1, Apo-2, and CD261.
It is expressed at low levels by activated T cells and some tumors. After TRAIL engagement, DR4 (TRAIL-R1), through activation of NF-κB, induces apoptosis in the TRAIL ligated cell.
The whole human DR4 genome was inserted at Hipp11 (H11) locus in B-hDR4 mice.
Human DR4 was detectable in homozygous B-hDR4 mice stimulated with anti-CD3ε in vivo but not in wild-type mice.

Protein expression analysis
Strain specific DR4 expression analysis in homozygous B-hDR4 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hDR4 mice (H/H) stimulated with or without anti-mCD3ε in vivo (7.5 μg/mice, stimulation for 24 hours, i.p.), and analyzed by flow cytometry with species-specific anti-DR4 antibody. Human DR4 was detectable in homozygous-stimulated B-hDR4 mice but not in wild-type mice.
