      Description
      • Lymphotoxin-β receptor (LTBR or TNFRSF3), a member of the tumor necrosis factor receptor superfamily (TNFRSF), serves as a receptor for both the LTA-LTB heterotrimer and TNFS14/LIGHT. Upon ligand binding, it activates NF-κB signaling and promotes apoptosis through TRAF3 and TRAF5, thereby playing a critical role in lymphoid organ development, immune responses, lipid metabolism, and programmed cell death.
      • The exons 1-7 of mouse Ltbr gene that encode signal peptide and extracellular domain are replaced by human counterparts in B-hLTBR mice. The genomic region of mouse Ltbr gene that encodes transmembrane and cytoplasmic portion is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are also retained. The chimeric LTBR expression is driven by endogenous mouse Ltbr promoter, while mouse Ltbr gene transcription and translation will be disrupted.
      • Human  LTBR was detectable on macrophages, DCs, neutrophils and monocytes from spleen, blood and bone marrow of B-hLTBR mice.
      • Application: This product is used for pharmacodynamics and safety evaluation of  tumors or autoimmune diseases.
      Targeting Strategy

Gene targeting strategy for B-hLTBR mice. The exons 1-7 of mouse Ltbr gene that encode signal peptide and extracellular domain are replaced by human counterparts in B-hLTBR mice.

Protein Expression Analysis on Dendritic Cells

Strain specific LTBR expression analysis in homozygous B-hLTBR mice by flow cytometry. Splenocytes, blood and bome marrow were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hLTBR mice(H/H), and analyzed by flow cytometry with species specific anti-mouse LTBR antibody (Biolegend, 113005) and anti-human LTBR antibody (Biolegend, 369903). Mouse LTBR was detectable in wild-type mice. Human LTBR was exclusively detectable in homozygous B-hLTBR mice.

      Protein Expression Analysis on Macrophages
