      Description

      CD7: A surface marker on T and NK cells, which can be used for targeted delivery.

      • Gene Information: CD7 is located on human chromosome 17. This gene encodes a transmembrane protein which is a member of the immunoglobulin superfamily.
      • Protein Expression: CD7 is Expressed on T-cells and natural killer (NK) cells and their precursors.
      • Pharmacological Mechanism: In vivo-generated, CD7-targeted CAR-T therapy works by delivering the CAR construct specifically to CD7-positive T cells and NK cells. This process directly engineers these host immune cells into functional CAR-T cells within the patient's body, which then eliminate tumor cells or autoreactive immune cells to treat associated cancers or autoimmune diseases.
      Targeting strategy

      CD7

      • Exons 1-4 of mouse CD7 gene that encode signal peptide, extracellular domain, transmembrane domain, cytoplasmic region are replaced by human counterparts in B-hCD7 mice.
      • The promoter and 5’ and 3’ UTR region of the mouse gene are retained. The human CD7 expression is driven by endogenous mouse CD7 promoter, while mouse CD7 gene transcription and translation will be disrupted.
      CD7 Protein Expression in Spleen
      • Human CD7 was detected on T, CD4+ T, CD8+ T and NK cells in B-hCD7 mice , but not in wild-type C57BL/6JNifdc mice.

      Human CD7 expression analysis in splenocytes. Splenocytes were collected from wild-type C57BL/6JNifdc mice, homozygous B-hCD7 mice. Human CD7 expression was analyzed by flow cytometry using species-specific anti-CD7 antibody ( biolegend, 343108).

      CD7 Protein Expression in Blood
      • Human CD7 was detected on T, CD4+ T, CD8+ T and NK cells in B-hCD7 mice , but not in wild-type C57BL/6JNifdc mice.

      Human CD7 expression analysis in blood. Blood was collected from wild-type C57BL/6JNifdc mice, homozygous B-hCD7 mice. Human CD7 expression was analyzed by flow cytometry using species-specific anti-CD7 antibody ( biolegend, 343108).

      Analysis of Leukocyte Subpopulations
      • The percentages of T cells, B cells, NK cells, DCs, neutrophils, monocytes, and macrophages in homozygous B-hCD7 mice are similar to those in C57BL/6JNifdc mice.
      • Humanization of CD7 does not affect normal immune cell development or splenic distribution.

      Analysis of leukocyte subpopulations by flow cytometry in immune organs and blood. Splenocytes, peripheral blood, and lymph nodes were isolated from female C57BL/6JNifdc and B-hCD7 mice (female, 9-week-old, n=3). Single live cells were gated on the CD45⁺ population and analyzed by flow cytometry as indicated. Values are expressed as mean ± SEM.

      Analysis of T Cell Subpopulations
      • The proportions of CD4⁺ T cells, CD8⁺ T cells, and Tregs in homozygous B-hCD7 mice are comparable to those in C57BL/6JNifdc mice.
      • Humanization of CD7 does not affect normal T cell development, differentiation, or splenic distribution.
