Soluble TL1A expression analysis in wild-type C57BL/6 mice and homozygous B-hTL1A/hPD-1 plus mice by ELISA. Bone marrow-derived dendritic cells (BMDCs) were produced by culturing bone marrow from wild-type C57BL/6 mice (+/+) and homozygous B-hTL1A/hPD-1 plus mice (H/H;H/H) and stimulated with LPS in vitro. After stimulation, cell culture supernatants were collected, and the levels of soluble TL1A were measured using a species-specific human TL1A ELISA kit. Soluble human TL1A was exclusively detectable in homozygous B-hTL1A/hPD-1 plus mice but not in wild-type C57BL/6 mice. Values are expressed as mean ± SEM. ND: not detectable.
Strain specific PD-1 expression analysis in wild-type C57BL/6 mice and homozygous B-hTL1A/hPD-1 plus mice by flow cytometry. Splenocytes were collected from wildtype C57BL/6 mice (+/+) and homozygous B hTL1A/hPD 1 plus mice (H/H;H/H), stimulated with anti CD3ε in vivo for 24 hours. The protein expression was analyzed with antimouse PD 1 antibody (Biolegend, 109104) and anti human PD 1 antibody (Biolegend, 329908) by flow cytometry. Mouse PD 1 was only detectable in wild type C57BL/6 mice.Human PD 1 was only detectable in homozygous B hTL1A/hPD 1 plus mice.
