We have generated a humanized mouse model expressing IL36R in place of the murine counterpart. mRNA and protein expression of human IL36R indicates successful generation of the humanized model. Humanization of IL36R does not change the overall development, differentiation or distribution of immune cell types in spleen, lymph node and blood. The anti-IL36R antibody Spesolimab analog reduced psoriasis-like skin lesions in B-hIL36R mice, indicating that B-hIL36R mice provide a powerful model for in vivo evaluation of anti-human IL36R antibodies.
